L929 cell line authentication

For biocompatibility assays and the cellasys #8 protocol we use the L929 cell line. As a prerequisition for reliability of data, our cell line was recently analysed by the Leibniz-Institut DSMZ-Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH according to the following procedure:

For the detection of human cells the DSMZ carried out DNA profiling using 8 different and highly polymorphic short tandem repeat (STR) loci. Furthermore, the sample was tested for presence of mitochondrial DNA sequences from rodent cells as rat, chinese and syrian hamster.

At a detection limit of 1:1000 no human STR loci sequences of the genomic DNA were detected. Additionally, at a detection limit of 1:100.000 no mitochondrial sequences from rat or chinese and syrian hamster cells were detected.

For verification of animal cell lines, the sample has been subjected to DNA Barcoding of Cytochrome Oxidase subunit 1 (COI) in order to identify the animal species.

The sample is derived of non-rodent but mouse, and non-human cell culture. Via COI DNA Barcoding the DSMZ revealed species-specific Mus muculus origin.

To achieve reproducible results, cellasys recommends to use the L929 cell line from DSMZ (DSMZ ACC 002).
